Endothelial cells line the inside of all blood vessels, forming a structurally and functionally heterogenous population of cells. Their complexity and diversity has long been recognized, yet very little is known about the molecules and regulatory mechanisms that mediate the heterogeneity of different endothelial cell populations. The constitutive organ‐ and microenvironment‐specific phenotype of endothelial cells controls internal body compartmentation, regulating the trafficking of circulating cells to distinct vascular beds. In contrast, surface molecules associated with the activated cytokine‐inducible endothelial phenotype play (...) a critical role in pathological conditions including inflammation, tumor angiogenesis, and wound healing. Differentiation of the endothelial cell phenotypes appears to follow similar mechanisms to the differentiation of hematopoietic cells, with the exception that endothelial cells maintain transdifferentiating competence. The present review offers a scheme of endothelial cell differentially expressed endothelial cell molecules as targets for directed therapeutic intervention. (shrink)

Ecological developmental biology (eco‐devo) explores the mechanistic relationships between the processes of individual development and environmental factors. Recent studies imply that some of these relationships have deep evolutionary origins, and may even pre‐date the divergences of the simplest extant animals, including cnidarians and sponges. Development of these early diverging metazoans is often sensitive to environmental factors, and these interactions occur in the context of conserved signaling pathways and mechanisms of tissue homeostasis whose detailed molecular logic remain elusive. Efficient methods for (...) transgenesis in cnidarians together with the ease of experimental manipulation in cnidarians and sponges make them ideal models for understanding causal relationships between environmental factors and developmental mechanisms. Here, we identify major questions at the interface between animal evolution and development and outline a road map for research aimed at identifying the mechanisms that link environmental factors to developmental mechanisms in early diverging metazoans.Also watch the Video Abstract. This paper explores the apologetic nature of Augustine’s Confessions. It first takes a brief look at Augustine’s intricate view of the relationship between faith and reason, in order to provide a background to his employment of apologetic elements throughout Confessions. Both positive and negative apologetic elements are examined throughout the paper. Some positive apologetic elements include Augustine’s presentation of the implied ontological argument, the cosmological argument, the teleological argument, the argument from the experience of beauty, and the demonstration of the (...) coherence of the Christian conception of God (including his explorations with time and creation). His negative apologetic elements include dealing with detractors from the Christian faith such as the Manichees and their objections. Although studies in recent years have focused on the autobiographical and religious experiential dimensions of Confessions, this paper seeks to demonstrate that the apologetic dimension is also foundational to Augustine’s text. It is important to realize that the apologetic nature of Confessions does not detract from its other natures (autobiographical and religious-experiential) since objects, ideas and persons can have more than one nature – as is true with the case of Christ and a number line. It is my hope to revive a forgotten aspect of Confessions that is quite pertinent to the Church today. (shrink)

Augustine’s teaching on the indissolubility of marriage profoundly influenced the Western Christian tradition on the matter of divorce and remarriage. Augustine famously insisted that while divorce was allowed in limited circumstances, remarriage was prohibited for both the guilty and the innocent parties. Less frequently acknowledged is the degree to which Augustine expressed doubt about the validity of his own teaching. In this essay I argue that even though Augustine offered a strict interpretation of the biblical evidence, he did so only (...) tentatively and often expressed doubts about the adequacy of his own views. The reason for this hesitation, I suggest, was Augustine’s knowledge that the meaning of the scriptural texts was ambiguous at best and that a significant portion of the previous tradition differed from the interpretation he favored. In the past 2 decades latent variable modeling has become a standard tool in the social sciences. In the same time period, traditional linear structural equation models have been extended to include non-linear interaction and quadratic effects (e.g., Klein and Moosbrugger, 2000 ), and multilevel modeling (Rabe-Hesketh et al., 2004 ). We present a general non-linear multilevel structural equation mixture model (GNM-SEMM) that combines recent semiparametric non-linear structural equation models (Kelava and Nagengast, 2012 ; Kelava et al., 2014 ) with (...) multilevel structural equation mixture models (Muthén and Asparouhov, 2009 ) for clustered and non-normally distributed data. The proposed approach allows for semiparametric relationships at the within and at the between levels. This work of vast erudition, which according to the author is substantially the same as the original Swedish edition, attempts to show how the young Augustine integrated certain key concepts of ancient philosophy into Christian thought. Holte limits himself to the philosophical issues and so does not consider the young Augustine as theologian. Parts I and II develop the historical background of the concept telos in the ancient philosophers and in the early Fathers. Part III presents the central thesis: that (...) Augustine tried to link the contemplative life to the commandment of love which requires total abandonment to God of every form of human life. The Telos is to be found in the contemplative life, although Augustine never separates it from the active. Telos is to be realized in the study of Christian philosophy and scripture. Unfortunately, today many scholars and general readers neglect the work of the Fathers of the Church. One often hears that the writings of the Fathers have become less relevant or less important because there has been so much progress in Scripture studies and because the Fathers did not really try to find out what the inspired writer meant to say-for they relied on allegory. St. Augustine's exegesis poses a particular difficulty in terms of its reliance on allegory. Augustine was strongly influenced by the allegorical exegesis of bishop Ambrose and recounts in his Confession.5 (VI, iv, 6) how he used to hear Ambrose teaching in his sermons, as though he were most diligently teaching a rule: "The letter kills, but the spirit gives life." Father de Margerie admits that Augustine did indeed suffer from "excessive allegorism" (126), but he insists that this is no reason to give up entirely on Augustine. Instead, with keen scholarly analysis he shows that, in spite of some defects, Augustine did in fact give us some really fine work in Scripture study. Father de Margerie stresses that according to Augustine the purpose of all Scripture is to teach love: "... the plenitude and the end of the Law and of BOOK REVIEWS 507 all the sacred Scriptures is the love... of a Being which is to be enjoyed and of a being that can share that enjoyment with us" (20; from De Doctrina Christiana, I, xxv, 39). This love is taught even at the beginning of Scripture: "... on a subjective level, Moses wrote for the love of all men" (30). But how could this be? "It seems to us nowadays that Augustine failed to perceive adequately the progressive nature of the disclosure of religious truths by the God who revealed them" (30). Father de Margerie responds to this objection by pointing out that given the belief that Moses had been given a transient, "face to face vision of the Divine being... and that this experience was granted him precisely in favor of the universal Church," it makes sense to assume that "the prophet and maker of the old Law received knowledge, during this vision and through divine revelation, of how future interpretation of the Law he was issuing-like that which Jesus was to issuewould extend even to enemies the love of one's neighbor" (31). Augustine was led to this view in the light of the varied interpretations being proposed in his own time for Genesis (48). Father de Margerie admits that "Augustine himself limits the scope of his theory concerning the plurality of literal meaning to a few rather exceptional verses of scripture" (65). Augustine believed, however, that Moses had received most remarkable gifts: I cannot think that Moses Your most faithful servant was given by you lesser gifts than I should have wished and longed to have for myself if I had been born at the same time as he....
